Intended use

Used for the cultivation, isolation, and enumeration of a wide variety of non-fastidious microorganisms in clinical, pharmaceutical, and diagnostic laboratories.

Principle

Formulated with enzymatic digest of protein and beef extract supplying essential amino acids, nitrogen, carbon, and vitamins, with high-clarity agar base ensuring optimal colony morphology.

Design

Pre-poured in optically clear, sterile polystyrene Petri dishes with uniform media depth to prevent drying and provide smooth streaking surface.

Nutrient Agar is a simple, general-purpose solid culture medium used for the cultivation, isolation, and maintenance of non-fastidious microorganisms. It is one of the most commonly used basal media in microbiology laboratories and serves as a foundational medium for routine culture work, teaching laboratories, and quality control applications. Due to its uncomplicated formulation and reliable performance, Nutrient Agar is widely employed for routine bacterial growth where no selective or differential properties are required.

Principle :
The principle of Nutrient Agar is based on providing a balanced supply of basic nutrients required for bacterial growth. The medium supplies nitrogen, carbon, vitamins, minerals, and salts in readily utilizable forms, thereby supporting the growth of a broad range of non-fastidious organisms. Since the medium does not contain inhibitory or indicator substances, it allows unhindered growth and natural colony development of organisms.

Composition :
Nutrient Agar typically contains peptone as a source of organic nitrogen, amino acids, and peptides; beef extract which provides water-soluble vitamins, carbohydrates, and growth factors; sodium chloride to maintain osmotic equilibrium; and agar as the solidifying agent. The formulation is carefully standardized to ensure consistent texture, clarity, and growth-supporting capacity.

Growth Characteristics :
The medium supports good growth of a wide variety of non-fastidious Gram-positive and Gram-negative bacteria. Colonies generally appear well defined and show characteristic morphology specific to the organism being cultured. The clear nature of the medium allows easy observation of colony size, shape, margin, elevation, and pigmentation.

Quality Control :
Each batch of Nutrient Agar is tested for sterility and growth promotion performance to ensure reliable and reproducible results in routine laboratory use.

Colony Morphology  :
Escherichia coli typically produces smooth, circular, moist colonies. Staphylococcus species form round, convex, opaque colonies that may appear creamy. Bacillus species often produce large, irregular, flat colonies with rough edges. Pseudomonas species may produce spreading colonies with characteristic pigmentation.

Intended Use :
Nutrient Agar is intended for use in routine cultivation, isolation, and maintenance of non-fastidious bacteria. It is also widely used for subculturing, purity checks, and preservation of laboratory strains.

Applications :
The medium is used in clinical laboratories for preliminary culture work, in pharmaceutical and food industries for quality control testing, in academic institutions for teaching and training purposes, and in research laboratories for general microbiological studies

Limitations :
The medium does not support the growth of fastidious organisms such as Haemophilus and Neisseria species. Occasional growth of Gram-positive organisms may occur if the inhibitory action is reduced or when heavily inoculated specimens are used.

Available Formats :

Storage and Shelf Life :
Prepared plates should be stored at 2–8°C and protected from light and dehydration. Shelf life: 60 days, as stated on the product label, valid under recommended conditions.

Handling and Precautions :
Plates should be brought to room temperature before inoculation to prevent condensation. Standard aseptic techniques must be followed during handling and inoculation to avoid contamination.
