MacConkey Agar (MACCONKEY AGAR W/O CV W/0.15% BILE SALT PLATES) is a selective and differential culture medium specifically designed for the isolation, identification, and differentiation of Gram-negative enteric bacteria. It is one of the most widely used primary plating media in routine bacteriology and plays a critical role in clinical diagnostics, food microbiology, pharmaceutical quality control, and water testing laboratories. The medium selectively suppresses Gram-positive organisms while allowing the growth of intestinal Gram-negative bacilli, making it highly valuable for the detection of enteric pathogens and coliform organisms.
Principle
MacConkey Agar is the earliest selective and differential medium for cultivation of enteric microorganisms from a variety of clinical specimens. Peptone and proteose peptone serve as a source of carbon, nitrogen, long chain amino acids, and other essential growth nutrients. The selective action of this medium is attributed to bile salts, which is inhibitory to most species of Gram-positive bacteria. Gram-negative bacteria usually grow well on the medium and are differentiated by their ability to ferment lactose. Lactose fermenting strains grow as red or pink colonies due to the production of acid from lactose, absorption of neutral red, and a subsequent colour change of the dye when the pH falls below 6.8. Lactose non-fermenting strains (such as Salmonella and Shigella) remain colourless and transparent.
Composition
The medium typically contains peptone and proteose peptone as sources of nitrogen, amino acids, and growth factors essential for bacterial metabolism. Lactose is included as the fermentable carbohydrate for differentiation. Bile salts (0.15%) act as selective agents inhibiting Gram-positive organisms. Sodium chloride maintains osmotic balance within the medium. Neutral red serves as the pH indicator, and agar is used as the solidifying agent. The formulation is carefully standardized to ensure consistent selectivity, differentiation, and colony morphology.
Colony Morphology & Differential Characteristics
- Escherichia coli: Typically produces flat to slightly raised, pink to red colonies with a smooth surface, often accompanied by a distinct zone of precipitated bile salts around colonies.
- Klebsiella species: Form large, mucoid, pink colonies due to prominent capsule formation.
- Enterobacter species: Produce raised, pink colonies.
- Salmonella & Shigella species: Form pale, colorless, transparent colonies without alteration of the surrounding medium.
- Proteus species: Produce colorless colonies with swarming waves effectively inhibited by the bile salts formulation.
- Yersinia enterocolitica: May appear as small, non-lactose fermenting colonies after incubation at room temperature.

Limitations
