Intended use

Intended for the isolation, cultivation, and preliminary differentiation of fastidious and non-fastidious bacterial pathogens from clinical specimens based on hemolytic activity.

Principle

Nutrient-rich base enriched with 5% sterile sheep blood providing vital growth factors and allowing clear visualization of alpha (greenish), beta (clear lysis), and gamma hemolysis.

Sheep Blood Agar Plate (Blood Agar Plates 5%) is an enriched, non-selective and differential culture medium widely used for the isolation and cultivation of a broad range of clinically significant microorganisms. It is one of the most important primary plating media in diagnostic microbiology laboratories and is extensively used for the recovery of fastidious and non-fastidious organisms from clinical specimens. Sheep Blood Agar Plate also allows the observation of hemolytic reactions, which provides valuable preliminary information for the identification of bacterial species, particularly streptococci and related organisms.

Principle

The principle of Sheep Blood Agar Plate is based on the enrichment of a nutrient agar base with sterile defibrinated sheep blood, typically at a concentration of 5%. The blood supplies additional growth factors, proteins, and nutrients required by fastidious organisms. Certain bacteria produce hemolysins that act on red blood cells present in the medium, resulting in characteristic hemolytic patterns. These hemolytic reactions are categorized as:

  • Alpha Hemolysis (α): Partial lysis of red blood cells producing a greenish or brownish discoloration around colonies due to hemoglobin reduction.
  • Beta Hemolysis (β): Complete lysis of red blood cells creating a distinct, clear, transparent zone surrounding colonies.
  • Gamma Hemolysis (γ): Absence of hemolysis with no visible change in the medium surrounding colonies.

Composition

Sheep Blood Agar Plate consists of a nutrient-rich agar base containing peptones, sodium chloride, and agar, supplemented with 5% sterile defibrinated sheep blood. The agar base provides essential nutrients required for bacterial growth, while the added blood enhances the growth of fastidious organisms and enables clear visualization of hemolytic activity. The formulation is carefully controlled to ensure optimal clarity, uniform distribution of blood, and consistent hemolytic reactions.

Colony Morphology on Blood Agar

  • Streptococcus pyogenes (Group A Strep): Small, translucent colonies surrounded by a wide, well-defined clear zone of beta (β) hemolysis.
  • Streptococcus pneumoniae: Small, flat or moist colonies with alpha (α) hemolysis (greenish halo), often showing a central depression (umbilication) with extended incubation.
  • Staphylococcus aureus: Medium-sized, convex, opaque golden-yellow or white colonies that may exhibit beta (β) hemolysis.
  • Enterococcus species: Small, compact, non-hemolytic (gamma γ) colonies.

Applications

The medium is routinely used in hospital diagnostic laboratories, reference laboratories, and academic institutions for primary isolation of pathogens from throat swabs, sputum, pus, wound swabs, blood cultures, and sterile body fluids; assessment of hemolytic reactions; and support of fastidious bacterial growth.

Limitations

The medium is non-selective and may support the growth of mixed flora, which can complicate interpretation in heavily contaminated specimens. Hemolytic patterns alone are not sufficient for definitive identification and must be supplemented with further biochemical or serological tests.

Quality Control & Performance Standards

Sheep Blood Agar is manufactured under stringent ISO 13485 quality control systems. Each batch is evaluated using reference strains (including Streptococcus pyogenes ATCC 19615 for beta hemolysis, Streptococcus pneumoniae ATCC 6305 for alpha hemolysis, and Staphylococcus aureus ATCC 25923) to ensure sterility, growth promotion, uniform blood distribution, and accurate hemolytic reactions.

Storage & Handling Precautions

Prepared Sheep Blood Agar plates should be stored refrigerated at 2–8°C in sealed packaging, protected from freezing and excess light. Shelf life is 60 days under recommended storage conditions. Plates should be brought to room temperature prior to use to prevent condensation. Care should be taken to maintain aseptic technique during inoculation.
